Plasmodesmal localization of the GHL17 protein PdBG1 m-Citrine fusion. Aniline blue staining of callose deposits (blue) and the green and blue channels superimposed are also shown (white arrows show co-localization). Scale bars = 20 um (from Gaudioso-Pedraza and Benitez-Alfonso, Frontiers in Plant Science, 2014).
Diffusion of GFP when expressed under the phloem companion cell promoter SUC2 in Arabidopsis wildtype root apical meristem and in the differentiation zone. Restricted diffusion of GFP in gat1 mutant is also shown. Scale bars = 20 um (Extracted from Benitez-Alfonso et al., PNAS, 2009 and Benitez-Alfonso et al., Dev Cell, 2013).
The figure shows a reconstructed tomogram section of a simple plasmodesmata found in a 6-day-old Arabidopsis root. The picture was taken by William Nicolas at the Bordeaux imaging Centre and was extracted from Knox and Benitez-Alfonso, COPS, 2014.
The figure shows wildtype, PdBG1OE and pdbg1,2 Arabidopsis roots expressing the reporter pDR5-3XVenus. PdBG are plasmodesmata- located beta glucanases. PdBG1OE are expressing PdBG1 fused with mCitrine (green cell walls). Wildtype and pdbg1,2 were counterstained with FM4-64 (red). Figures were extracted from Benitez-Alfonso et al., Dev Cell, 2013.
Immunolocalization of callose (green) in the differentiation zone of Arabidopsis root stained with DAPI (cyan). Lateral root primordia (LRP), xylem pole pericycle (XPP), endodermis (E) and cortex (C) are indicated. Black arrows show PD sites. Figure extracted from Benitez-Alfonso et al., Dev Cell, 2013. Protocol will appear in Pendle and Benitez-Alfonso, Method Mol Biol, 2015.
Time-lapse movie of the development of Tobacco rattle virus (TRV) infection in Nicotiana benthamiana as revealed from the virus-driven expression of green fluorescent protein (GFP) (published with permission from the John Innes Centre).
